چکیده

Equations 2 and 3, came from investigations with a highly purified enzyme from Micrococcus aerogenes (6), and from experiments with the pigeon liver enzyme (7-9). On the other hand, data (10, 11) obtained with a formate-activating enzyme from Clostridium cylindrosporum have led to an alternate interpretation, namely that Reaction 1 goes via a concerted mechanism without the formation of any detectable intermediates. The present communication provides further information defining the over-all reaction (Equation 1) catalyzed by the M. aerogenes enzyme. A two-step mechanism is presented which modifies Equations 2 and 3 to include the role of the enzyme.

I. Isolation and Crystallization of the Enzyme?

Greenberg and associates (2, 3) in pigeon liver homogenates and was later found in a variety of bacterial species, including Clostridium cylindrosporum (4), Micrococcus aerogenes (5), and others (6). The enzyme has been purified from acetone powders of pigeon liver (7), from Micrococcus aerogenes (6), and from Clostridium cylindrosporum (8, 9). This paper describes the purification of the enzym...

Preparation and properties of cell-free "formic hydrogenlyase" from escherichia coli.

The available evidence indicates that formic acid is the precursor of the molecular hydrogen produced as an end product of fermentations effected by Escherichia coli, Aerobacter aerogenes, and related organisms. Formate is rapidly decomposed by these bacteria with the formation of H2 and CO2 in a reaction catalyzed by the enzyme or enzyme-complex designated as "formic hydrogenlyase". One of the...

Formic Acid Activation in Plants. I. Purification, Properties and Distribution of Formyltetrahydrofolate Synthetase.

Tetrahydrofolic acid derivatives have been found to be the active cofactors in a number of reactions which involve 1-carbon moieties (9). The participation of tetrahydrofolate enzyme systems in purine and histidine biosynthesis has been well established (9). N10-formyltetrahydrofolate apparently functions in purine synthesis by serving as a 1-carbon donor (2, 9). The synthesis of N10-fornlyltet...

Formic Acid Activation in Plants. II. Activation of Formyltetrahydrofolate Synthetase by Magnesium, Potassium, and Other Univalent Cations.

The purification, properties and distribution of the formate activating enzyme, formyltetrahydrofolate synthetase, from plants were described in the preceding paper (5). Studies of the activation of the enzyme by magnesium and univalent cations are described in this paper. The mechanism of action of magnesium in biological catalysis is reasonably well understood. Most transphosphorylation react...
